Calcium measurement in Melanocytes

Normal Human Epidermal Melanocytes (NHEM), isolated from one adult donor. The donor is a Caucasian woman, 33 years old, with an estimated skin phototype of II/III.

Endothelin-1 was tested at 5 concentrations (1 nM to 100 nM) to assess the ability of the NHEM to respond via an endogenous G Protein Coupled Receptor (GPCR) in a calcium assay.

DC and LC play a key role in the induction phase of contact allergenicity and other immunological reactions of the body. In skin infections, the local Langerhans cells take up and process microbial antigens to become fully functional antigen-presenting cells.

Evaluation CGRP receptors antagonists

A functional assay assessing antagonistic properties of compounds on endogenous CGRP receptors, activated with the potent vasodilator CGRP (Calcitonin Gene-Related Peptide). Primary Human Dermal Microvascular Endothelial Cells were seeded in 96-well plates. Since CGRP receptors are Gs-coupled GPCRs, cAMP accumulation was quantitated, by HTRF. The dose-dependent aCGRP-induced cAMP production was rightward-shifted by the reference antagonist CGRP 8-37, indicating inhibition.
